In particular, his team discovered that CTLA4 is a negative regulator of T cell activation (cited 2,000 times), paving the way for the development of anti-CTLA4 agents now in clinical use for autoimmune diseases. “My decision to stay in Canada motivated me to assemble a group of outstanding doctors and researchers at the Princess Margaret Cancer Centre to make concrete advances in the fight against cancer,” he says.
As a senior scientist with the Princess Margaret Cancer Centre and director of the Campbell Family Institute for Breast Cancer Research since 2004, Mak has earned international recognition and has even been named to the Canadian Medical Hall of Fame.
Mak is a classic example of a brilliant international student who has made his adopted country better by calling it home.
In particular, he is interested in mechanisms by which tumor cells undergo metabolic adaptation and control genomic instability, with the goal of identifying potential targets for novel cancer therapeutics.
Professor Mak has published over 800 peer-reviewed research papers and holds many patents. He is best known as the lead scientist of the group that first cloned the genes of the human T cell antigen receptor, a discovery that provided essential insights into the molecular basis of cellular immunity.

His lab produced numerous important studies, which have been cited a record 40,000+ times.The next two decades saw Mak blaze a trail in the understanding of the molecular biology of the immune system and cancer at the cellular level.
Dr. Mak’s lab also made major contributions to defining the functions of PTEN (cited more than 2,000 times) as well as the relationship between the breast cancer susceptibility genes BRCA1 and BRCA2 and defects in DNA repair.
